We found that RB and SUCLA2 co-deletion was frequently observed in primary and advanced prostate adenocarcinoma patients. Using SUCLA2 knockout and reconstituted DU145 cells, we performed chemical screening with natural compounds and identified thymoquinone that specifically kill SUCLA2 deleted cells. To elucidate molecular mechanism, we employed cap analysis gene expression-sequence as a discovery platform to identify gene set of SUCLA2 deletion with thymoquinone treatment. We compared four groups which are SUCLA2 knock out or reconstituted cells treated with DMSO or thymoquinone.
