The development of oligodendrocytes, the myelin-forming cells, is crucial for the normal functioning of the central nervous system. The mechanisms underlying this process, particularly the role of alternative splicing, remain largely unelucidated. Here, we report that the absence of splicing factor SRSF10 in oligodendrocyte lineage cells (OLCs) significantly impairs myelin formation and reduces the number of oligodendrocytes in the mouse central nervous system.By combining the results from RIP-seq and RNA-seq, we identified a series of genes with altered alternative splicing patterns that are directly regulated by SRSF10. Our findings have significant implications for understanding the development of oligodendrocytes and exploring potential therapeutic approaches for white matter-related diseases.
