Patient-derived lymphomoids preserve the tumor architecture and allow testing response to therapies in lymphoma [scRNA-seq]

We designed an ex vivo system to culture tissue explants from lymphoma tissue. To determine if the explants derived from vavP-Bcl2 mouse spleens retained the cellular composition and transcriptional profile of the cells in the original tissue, we analyzed six lymphomoids derived from the spleens of two mice, after one or two weeks in culture, and the original spleen tissues by single-cell RNA-sequencing. Overall design: Two lymph node biopsies were used for this analysis. One biopsy was taken from a patient with DLBCL (patient 08 in this study) and a second biopsy contained reactive lymphoid tissue ( (patient 03 in this study). In addition, for patient 08, we analysed tissue explants grown for 3 days untreated or treated with 2 mM ibrutinib or 10 mM idelalisib.
