To identify metabolic pathways that are essential for the progression of TNBC in vivo 4T1 breast cancer cells were engineered to express synergistic activation mediators. Then these cells were transfected with an sgRNA library containing 69,225 sgRNAs targeting 23,439 protein-coding genes and 491 control sgRNAs.These tumor cells were transplanted into mice. The tumors on days 5 and 10 were collected for sgRNA sequencing.
