Cytogenomic and phylogenomic evidence for new infrageneric relationships in Macroptilium (Benth.) beans

Our aim was to perform a cytogenomic comparative analyses in Macroptilium species using the available oligo probes from P. vulgaris and V. unguiculata, as well as 5S and 35S rDNA markers, to identify and characterize the karyotype pattern for the genus and investigate the intergeneric and interspecific variation. We also sequencing the genome of two species in low coverage, to investigate in silico the proprieties of repetitive fraction dynamic inside Macroptilium. We were able to determine the oligo-FISH pattern, which showed high interspecific and intergeneric synteny, even with APK. Although Macroptilium presented independent translocations, variation in rDNA sites and different abundance of satellites DNA, which do not support the actual phylogenetic distribution of the species but allowed the creation of three cytogenetical groups. Altogether, we demonstrate the implementation of different cytogenomic tools to characterize and create cytogenetic groups for a closely related genus from bean crops.
