The pER:MYB48 and pER:bHLH68 expression construct were transfected into Arabidopsis stem. Arabidopsis stems after bolting 15 days were treated with 10 mmol/L estradiol for 2 h, 4 h, 6 h and 8 h, respectively, and DMSO treatment was used as control. After treatments, the stems were harvested and used for total RNA isolation and RT-qPCR and transcriptional analyses. pER:MYB48 and pER:bHLH68 materials were selected after estrogen induction, and total RNA was extracted for transcriptome sequencing.
