The data set and scripts are Supplemental Material data for the manuscript 'Fluorescence Lifetime Imaging Microscopy (FLIM) visualizes internalization and biological impact of nanoplastics in live intestinal organoids ' by Irina A. Okkelman, Hang Zhou, Sergey M. Borisov, Angela C. Debruyne, Austin E. Y. T. Lefebvre, Marcelo Leomil Zoccoler, Linglong Chen, Bert Devriendt, Ruslan I. Dmitriev. The data set attached is a number of raw ptu files of life imaging FLIM of pig intestinal organoids from the control ('auto') group and loaded with individual D and B nanoparticle species and their 1:1 mixture and the corresponding csv files of GS phasor coordinates from individual organoids. The attached Python-based scripts (txt files) can be used for the postprocessing of the data set accordingly:
Supplementary code_ area_GS_export - napari macros for analysis of ptu microscopy imaging files; determines and exports a ROI square area (csv files) and ROI phasor plot GS coordinates (csv files) from phasor plots reconstructed in napari-flim-phasor-plotter with binning 3, harmonic 1, threshold 188, frequency pulse 80 MHz settings. Supplementary code_GS_count - filtration of nonsense (value 0) GS coordinate events from the list of GS phasor coordinates (csv files) and counting of the total valid GS events
Supplementary code_tau_zones - can be applied only to the corresponding experimental csv data set, as it utilizes predetermined parameters of the average fingerprint GS coordinates from the exact data set. The code can be used to count the percentage of phasor events in lifetime zones: zone D( pure nanoparticles D), zone B (pure nanoparticles B) and zone D/B (co-detected D and B nanoparticles).
