Human embryonic stem cells (hESCs) are naturally equipped to maintain genome integrity, which minimizes genetic mutations during early embryo development. However, during in vitro culture, the risk of genetic aberrations and subsequent cellular changes in hESCs threatens stem cell therapy. While a few studies have reported specific somatic mutations and copy number variations, the molecular mechanisms underlying culture-adapted phenotypic changes in hESCs are not well-understood.Therefore, we conducted comprehensive genomic and single-cell transcriptomic analyses on an isogenic hESC model that displayed definitive culture-adapted phenotypes. Additionally, the whole-genome and transcriptome of the hESC lacking TP53 were analyzed.
