To systematically identify the biological processes and signaling pathways governed by G-alpha-protein-encoding gene SsGpa1 involving vegetative incompatibility response in Sclerotinia sclerotiorum, we performed transcriptomic analysis on strain ?SsGpa1-2 and its parental strain Ep-1PNA367, respectively, when they were dual-cultured with either incompatible strain 1980 or compatible strain Ep-1PNA367. Dual-culture assay was performed by partner strains that was placed opposite on cellophane-overlaid PDA until the hyphae of two colonies grow into each other and interact for 6 hours. Mycelium samples from the contact zone (0.5-1.5 cm) of three plates were pooled per replicate, and total RNA was extracted using Trizol regent. Sequencing was performed on an Illumina HiSeq 4000 (Bioyi Biotechnology, Wuhan, China), generating approximately 6 GB of raw data per sample.
