Tissue resident memory T (TRM) cells mediate the frontline protection against melanoma lung metastasis. We utilized scRNA-seq to investigate the underlying molecular mechanism of central memory T (TCM) to lung TRM development in the prime-boost vaccinaiton regimen. Overall design: We immunized groups of C57BL/6 mice with DNA vaccine DNA-OVA i.m./EP plus live-attenuated influenza-vectored vaccine LAIV-HK68-OVA i.n. regimen and extracted total cells from lungs, MLN, and spleen at 21 days post DNA-OVA prime immunization (termed Day 0), 2 days, 4 days, and 30 days post LAIV-HK68-OVA booster vaccination for single-cell RNA sequencing (scRNA-seq).
