TransMPRA sequencing data.

Gene regulation occurs through trans-acting factors (e.g. transcription factors) acting on cis-regulatory elements (e.g. enhancers). Massively parallel reporter assays (MPRAs) functionally survey large numbers of cis-regulatory elements for regulatory potential, but do not identify the trans-acting factors that mediate any observed effects. Here we include preliminary data from a pilot transMPRA experiment — a reporter assay that efficiently combines multiplex CRISPR-mediated perturbation and MPRAs to identify trans-acting factors that modulate the regulatory activity of specific enhancers. Overall design: 2 spike-in fastq files with information used to associate random barcodes that identify specific gRNAs. 24 fastqs with RNA or DNA sequencing information of reads from the 2323 distinct transMPRA constructs containing many distinct barcodes. These were collected from 3 replicates at 2 library concentrations (lowMOI vs highMOI) from cells harvested at 2 time points (day 5 vs day 10).
