Knockdown of HP1γ in H1792 cells

For analysis of genes regulated by HP1γ in LUAD, we used microarray analysis to determine the global changes in gene expression upon HP1γ knockdown in H1792 cells and identified distinct sets of up-regulated and down-regulated genes upon depletion of HP1γ. H1792 cells were infected with 2 different shRNAs of HP1γ (shHP1γ-16 and -17). After 96 hours of selection in 1ug/ml puromycin-containing medium, cells were harvested for RNA extraction. Global gene expression levels of HP1γ knockdown cells were measured by Affymetrix U133P and compared with those of control (shLuc) cells. Experiments were peformed in duplicate. Microarray adat was processed by RAMA and heat map was generated by MeV software.
