The cellular diversity of stromal compartment within the tumor microenvironment might change dynamically as tumors evolve that allowing distant dissemination of tumor cells and informing the treatment options. Thus, we used single cell RNA sequencing (scRNA-seq) to provide a detailed insight of cellular components feature in bladder cancer. Fresh bladder cancer tissues and normal adjacent tissues were obtained from patients and washed with ice-cold PBS. Then, the tissues were subjected to prepare single cell suspensions with tumor dissociation kit. Subsequently, the single cell suspensions were used to generate single-cell Gel Bead-In-EMlusion (GEMs) by a 10X Genomics GemCode Single-cell instrument and construct the cDNA libraries. The library sequencing was performed on the Illumina novaseq 6000.
