Primary lung fibroblasts were isolated from C57BL/6 mice by enzymatic digestion and confirmed by immunofluorescence staining to be>=95 % pure. Cells were then exposed to 10 ng/mL TGF-beta for 12 hours to generate a fibrotic model. Iso-seq was subsequently used to analyze the full-length transcriptome response of these fibroblasts to TGF-beta stimulation.
