Tumor Genome and Microenvironment Alteration by Trastuzumab Deruxtecan as Neoadjuvant Therapy for HER2 Mutant NSCLC

In patients with locally advanced non-small cell lung cancer (NSCLC) harboring HER2 exon 20 insertions, we administered neoadjuvant T-DXd therapy. Tumor tissues and adjacent normal tissues were collected before and after treatment for whole-genome sequencing. In addition, post-treatment tumor samples, as well as near-adjacent (3 cm from the tumor margin) and far-adjacent (15 cm from the tumor margin) tissues, were obtained for transcriptome sequencing, with three biological replicates per sample type.
