CONTRAST D4.3 dataset: eDNA metabarcoding ASV tables (16S and 18S)

CONTRAST D4.3 Dataset – README Dataset descriptionThis dataset contains Amplicon Sequence Variant (ASV) tables generated from 16S and 18S rRNA gene metabarcoding of marine sediment samples collected within Work Package 4 (WP4) of the CONTRAST project. The data originate from a mesocosm experiment investigating the effects of the contaminant of emerging concern (CEC) citalopram on benthic microbial and meiobenthic communities. Files included- citalopram_16s-asv-table.csv ASV table (16S rRNA gene) containing sequence variants and associated taxonomic assignments. Values represent read counts per sample. - citalopram_18s-asv-table.xlsx ASV table (18S rRNA gene) containing sequence variants and associated taxonomic assignments. Values represent read counts per sample. - citalopram_18S_16S-metadata.csv Combined metadata file for both 16S and 18S datasets. The column “marker” indicates whether each sample corresponds to the 16S or 18S dataset. Data structure- Rows in ASV tables represent individual ASVs- Columns represent samples- Entries correspond to read counts per ASV per sample- Metadata links samples to experimental conditions Experimental design (summary)Samples include:- Field reference samples- Initial baseline samples (T0)- Controls (blank and solvent control)- Exposure treatments across a gradient of citalopram concentrations ProcessingData were generated using standardised environmental DNA (eDNA) workflows developed under Task 4.1, including:- DNA extraction from sediment samples- 16S and 18S rRNA gene amplicon sequencing- Bioinformatic processing based on ASVs- Taxonomic assignment using public reference databases Notes- This dataset represents full ASV tables (read counts) used to derive summary outputs presented in Deliverable D4.3.- Presence–absence summaries and “top taxa” tables shown in the report are derived from this dataset.- A single metadata file is provided for both markers; the “marker” column specifies the sequencing target (16S or 18S).
