In this study, we generated human pluripotent stem cell-derived myogenic progenitor cells and transplanted into injured skeletal muscle. We performed two single cell RNA-seq experiments. In the first experiment, we compared transplanted satellite cells with in vitro controls. In the second study, we did a time-series analysis of transplanted cells. Overall design: In the first experiment, we sequenced 1) human embryonic stem cells, 2) hESC-derived myogenic cells in vitro, 3) hESC-derived myogenic cells, cultured for a further month in vitro, 4) hESC-derived myogenic cells, transplanted in vivo and harvested one month later. In the second experiment, we sequenced hESC-derived myogenic cells, transplanted in vivo, at 1, 2, 3, and 4 weeks post-transplantation.
