Based on the Polysome profling method, a single concentration of sucrose solution was used as a buffer to separate ribosomal fractions from free mRNA and other cellular components in A498 cells by ultrafast cryo-centrifugation, and all the information of full-length mRNAs that were being translated could be obtained by combining high-throughput sequencing with the extraction of RNAs in the precipitates.
