Abstract:Duck hepatitis A virus 1 (DHAV-1) is a highly lethal virus that severely affected the duck industry worldwide. Long non-coding RNAs (lncRNAs) play nonnegligible roles in pathogen attack. However, the function of ducks lncRNAs during DHAV-1 insults remains unclear. Here, we conducted transcriptome sequencing to investigate the dynamic changes of host lncRNAs profiles in duck embryo fibroblasts (DEFs) infected with DHAV-1. Totally, 16589 lncRNAs were identified and genomic features were characterized. Additionally, 1908 and 1105 different expression lncRNAs (DELs) were screened at 12 and 24 hours post-infection (hpi). Moreover, we predicted the target genes to interact with DELs based on cis- and trans- regulatory manners and constructed lncRNA-target genes regulatory network. Finally, functional annotation analyses indicated putative target genes of DELs mainly participate in immune-related pathways, including the pattern recognition receptors (TLR3, DDX58, MDA5, LGP2, cGAS), signal transducer (STAT1), transcription factors, and effector molecular (IL6, IL8, IL10, TRIM25, TRIM35, TRIM60, IFITM1, IFITM3, IFITM5) and we confirmed the up-regulation of these target genes by qRT-PCR. Our study comprehensively analyzed the lncRNA profiles upon DHAV-1 infection and screened the target genes involved in innate immunity response, thereby revealing the essential roles of ducks lncRNAs and broadening our understanding of host-virus interactions.
