The research focused on the total chemical synthesis of High Mobility Group Box 1(HMGB1) protein and the biological effect of post-translational modifications. There are two synthetic routes developed separately for fully reduced HMGB1 and disulfide HMGB1. Sialylated complex type N-glycan was intalled to the fully reduced HMGB1 molecular and homogeneous glycosylated HMGB1 variants were synthesized to study their biological effects on receptor binding and cell chemotaxis. Acetylation was installed to both the fully reduced HMGB1 and disulfide HMGB1 to obtain homogeneous acetylated HMGB1 variants to study their biological effects on receptor binding and proinflammatory effects.
