Double mutant heat labile (dmLT) adjuvant is being proposed for oral vaccine formulation. Given that earlier experiments demonstrated dmLT instability in stomach acid and low pH an oral antacid buffer will be required for oral administration of dmLT. In general antacid buffers contain high salt concentrations. In order to limit the overall volume of antacid buffer required with a candidate vaccine dmLT stability in buffers with high osmolarity was investigated by ELISA. dmLT samples were incubated with buffers with high ionic strength and incubated at room temperature. Samples were taken at the specified time points and measured for dmLT stability by ELISA.
