We describe a next-generation sequencing (NGS)-based, high throughput screening approach for the identification of mycobacteriophage-encoded proteins that are toxic to mycobacteria. A plasmid derived library representing the mycobacteriophage TM4 genome was constructed and transformed into Escherichia coli and Mycobacterium smegmatis.
