Single-nucleus and single-cell RNA-sequencing of human pancreatic islets

Transcriptomics is frequently used to interrogate alterations in cultured human islet cells using single-cell RNA-sequencing. We introduce single-nucleus RNA-sequencing as an alternative approach for investigating human islets. The Nuclei EZ protocol was used to obtain nuclear preparations from human islet cells. Such preparations were used to generate single-nucleus RNA-seq datasets and compared to single-cell RNA-seq output obtained from cells from the same donor.
