Compare transcriptome between P6 and P30 cardiomyocytes in mouse. Overall design: Rosa-Tom or Rosa-Cas9 mice were injected with AAV9-cTNT-Cre at P1. cardiomyoytes were isolated at P6 or P30. In each experiment, cardiomyocytes were isolated by retrograde collagenase perfusion and purified by fluorescence activated cell sorting. Total RNA was extracted from FACS sorted cells. RNA-seq libraries were generated using SMART-seq and Nextera kits and were sequenced on a NextSeq500 sequencer. FASTQ Data were aligned by STAR and count matrix generated by FeatureCounts.
