We set out to determine how ploidy can impact the outcome of evolution, as well as the likelihood of evolutionary rescue, using short-term experiments with yeast (Saccharomyces cerevisiae) in a high concentration of the fungicide nystatin. We found no reliable diploid resistant mutants but many haploid mutants. These samples were sequenced as potential diploid resistant mutants, but all except 4_B11_D were determined to carry haploid contaminants, and 4_B11_D was not reliably resistant. All other sequenced samples except 1_F9_D are haploid, and 1_F9_D was found to carry haploid contamination in follow-up assays.
