To establish the role of the Glutamate Receptor-like gene in Marchantia polymorpha, MpGLR , in wound-induced gene expression changes, we compared the transcriptomes of plants that are wild type for MpGLR to the transcriptomes of Mpglr-7, a deletion mutant in MpGLR, analyzing samples from intact plants and from wounded plants, both from damaged tissues and from systemic undamaged tissues of wounded plants. Overall design: To investigate the function of MpGLR in wound-induced gene expression changes, we generated the CRISPR-cas deletion mutant Mpglr-7. We then performed gene expression profiling analysis using data obtained from RNA-seq of intact plants and of wounded plants, both from damaged tissues and from systemic undamaged tissues of those wounded plants. Samples were collected 2 hours after wounding the plants, at which time samples from intact plants were also collected. Plants were wounded by crushing two thallus lobes with serrated forceps (wounded tissues) while the adjacent undamaged lobes were collected as distal non-damaged tissues (systemic tissues). Three independent biological replicates of each sample were sequenced. Every independent biological replicate contained tissues from 8 plants.
