X-ray structures of Drosophila dopamine transporter in complex with nisoxetine and reboxetine.

Epitope residues were calculated from [PDB: 4XNU and 4XNX] as the antigen residues at 4 Å atomic distance from the antibody. The structure in PDB: 4XNX lacks contact residues H338, Y498, M510 and R598 and contains contact residue D599 not found in PDB: 4XNU. The epitope of 9D5 Fab on the dopamine transporter with the inhibitor nisoxetine bound was determined from the crystal structure of the complex, solved by molecular replacement. The Fab was used as a chaperone to aid in crystallization. The dopamine transporter construct dDAT has deletions of residues 1-20 and 164-206 and point mutations V74A, V275A, V311A, L415A and G538L to enhance thermostability.
