Gene expression profile at single cell levels from glomerulus in wild type (WT) and inhibitor of MyoD family A(I-mfa) knock out mice

Background: Inhibitor of MyoD family A (I-mfa) is a cytosolic protein and its function in kidney is unknown. In this study we used scRNAseq technique to study the diversity at single cell levels in kidney glomerulus from both wild types and I-mfa knockout mice seperately. Translational Project Award from American Heart Association (20TPA35500045, to R. Ma) Overall design: 4 Wild type (WT) mice and 4 I-mfa knock out (I-mfa KO) mice (12-13 weeks) were used for the study. All kidneys from WT mice and KO mice were pooled, respectively for sample preparation so that sufficient cell numbers could be obtained for single cell analysis. Single cells from kidney glomerulus in each mouse were isolated. Cell number count in the final cell suspension was achieved using a hemocytometer. Additionally, cell viability (>80% for both groups) was examined and determined using trypan blue staining. The cell suspension from both the WT and KO groups were further evenly divided into 3 aliquots of 100 µl each with a concentration of 600 cells/µl and 900 cells/µl, respectively. single cells in each sample were analyzed by scRNAseq.
