Single cell gene expression analysis of hematopoietic progenitors in Ddx41-heterozygous mice

To determine the effect of heterozygosity of Ddx41 on hematopoiesis, we conducted single cell transcriptome analysis on hematopoietic progenitor cells from Ddx41-heterozygous and control bone marrow. We found that heterozygoisty of Ddx41 did not signficantly alter the distribution of progenitor cell types, and changes in gene expression within the clusters were minimal. Bone marrow mononuclear cells from Ddx41 flox/+ or control mice were transplanted into lethally-irradiated recipient mice, and then the recipients were treated with tamoxifen to excise the floxed alleles. The bone marrow of these mice was harvested 8-weeks post-transplant and the Lineage-negative, cKit-psotive progenitor cells were sorted. After cell hashing to enable multiplexing, these cells were processed for 10X Chromium analysis.
