tRNA modifications play important role in regulating mRNA translation via codon decoding regulation. tRNA queuosine modification (tRNA-Q) is synthesized by the TGT complex, composed of QTRT1 and QTRT2 enzymes. The TGT complex utilizes queuine to convert guanosine in GUN anticodons to queuosine. tRNA-Q expands the codon recognition of GUN anticodons from NAC to NAC and NAU codons. Here, we developed QTRT1 and QTRT2 Knockout (KO) HEK293T cell lines to study the impact of tRNA-Q depletion on mRNA expression and translation using RNA-seq and Ribo-seq analysis.
