To assess the impact of B cell deficiency on MCMV-specific CD8? T cells, equal numbers of M45 tetramer-positive CD8? T cells were sorted from wild-type and B cellâdeficient JHT mice. The isolated antigen-specific CD8? T cells were subjected to bulk RNA sequencing to compare transcriptional profiles between the two groups. Overall design: Wild-type and B cellâdeficient JHT mice were infected with murine cytomegalovirus (MCMV). At 7 days post infection, MCMV-specific M45 tetramer-positive CD8? T cells were isolated by flow cytometric sorting. Equal numbers of tetramer-positive CD8? T cells from each group were collected to control for differences in cell frequency. Total RNA was extracted from the sorted cells and subjected to bulk RNA sequencing to compare transcriptional profiles between wild-type and JHT mice.
