A recombinant AAV9-SEAP vector batch was produced in a 50l bioreactor in HEK293 suspension cells. The viral DNA was extracted and converted to double stranded DNA by annealing, DNA polymerase I and reverse transcriptase. The samples were prepared and sequenced with the ONT V9 chemistry or the V14 chemistry. As comparison an annealed sample was sequenced using SMRT sequencing.
