We collected the plaque samples from 10 periodontitis patients and 11 periodontal healthy volunteers, cultivated under the selective pressure of the four antibiotics (amoxicillin, metronidazole, clindamycin and tetracycline) and performed microbiome analysis. Tetracycline-sensitive and resistant strains then were isolated and the expression of tetM gene was calculated by quantitative real-time polymerase chain reaction (q-PCR). In situ plaque culture first showed the oral microbiota from periodontitis patients were more resistant to antibiotics than that from healthy people. There were significantly differences for the microbial diversity and community structure between pre- and post- antibiotics treated in periodontitis groups, especially to amoxicillin and tetracycline. Metagenomic analysis, strain isolation and minimum inhibitory concentration further indicated a TCY-resistance gene tetM may contribute to the heterogeneous drug resistance to tetracycline in Streptococcus. The expression of tetM was found to be promoted by tetracycline in tetracycline-resistant isolates.
