Study of ebola virus VP35 protein on gene expression in HepG2 cells

The overall effect of VP35 on host gene expression were investigated by the differentially expressed genes (DEG) analysis in cells transfected with or without Flag-VP35 Gene expression in HepG2 cells transfected with VP35 or vector was analyzed with a gene expression microarray HepG2 cells were transfected with the plasmids Flag-VP35 or Flag, and each group has three independent replicates. Forty-eight hours later, total RNA from six samples was extracted using the RNeasy mini kit (QIAgen, USA) according to the manufacturer’s protocol. After first-strand cDNA synthesis with Poly-dT primer and second-strand cDNA synthesis, the cDNA was subjected to Gene Expression Microarray analysis following the manufacturer’s protocol. The chips were scanned with a GeneChip Scanner G2565CA (Agilent).
