Traditional mulberry leaf feeding, while nutritionally optimized through co-evolution, faces challenges such as seasonal limitations and pathogen contamination, prompting the adoption of artificial diets for year-round, industrialized sericulture. Despite significant advances in improving artificial diet formulations for sericulture in recent years, artificial diet -fed silkworms still suffer from problems such as reduced immunity, stunted growth and low larval survival rates. Notably, transcriptomic analyses highlight a marked downregulation of PGRP and AMP genes in artificial diet-fed silkworms, indicating impaired pathogen surveillance and immune activation. In this study, the role of midgut-specific BmPGRP-L1 in intestinal immunity was studied at the individual level by genetic manipulation, and the effect of its expressional changes on intestinal flora of silkworm reared on artificial diet was explored.
