To get insights into the molecular characteristics of Cspg4+ microglia, we crossed NG2-enhanced green fluorescent protein (EGFP) transgenic (Tg) reporter mice, expressing EGFP under the control of NG2 promoter, with A30P Tg mice, to generate A30P a-syn;NG2-EGFP double Tg mouse line. This double Tg mouse line allowed to analyze the responses of Cspg4+ cell populations labelled by EGFP, including microglia, following exposure to A30P a-syn. EGFP-labelled Cspg4+ cells were sorted by FACS and then subjected to single-cell RNA sequencing (scRNA seq).
