In this study, we unveil that transcriptionally distinct oligodendrocyte populations have spatial preference and differential response to spinal cord injury. We also assess by different methods, including single-cell RNA sequencing, that the developmental origin of oligodendrocyte progenitors does not determine their specification into mature olgodendrocyte populations. Overall design: We pooled the corpus callosum from two adult (P60) Emx1::Cre-Sox10::Cre-GFP-STOP-TdTom mice from which we took two samples (GFP+ and TdTom+ oligodendrocytes) to address the influence of the oligodendrocyte developmental origin in their subpopulation identity in adulthood. The corpus callosum of P60 Emx1::Cre-Sox10::Cre-LoxP-GFP-STOP-TdTom was sectioned at the microtome and microdissected from two mice. The injury site, rostral and caudal wallerian degeneration regions or control spinal cord of three months Sox10::Cre-GFP mice were microdissected from eight injured and five laminectomy mice.
