Mapping the IMiD-dependent cereblon interactome using BioID-proximity labelling

This project employs the bacterial biotin-ligase, BioID2, to biotinylate and purify proteins that interact with cereblon (CRBN), including those that interact in a transient and/or weak fashion. In this project, cells expressing the fusion protein BioID2-CRBN are exposed to IMiDs (immunomodulatory imide drugs) and/or a proteasome inhibitor (bortezomib), and proteins that are biotinylated are subsequently purified and processed for mass spectrometry. Proteomic analysis identifies proteins that are purified in the different conditions, allowing to define how the CRBN interactome changes in the presence of these commonly used anti-myeloma drugs.
