Purpose: To explore the mechanisms by which PRDM16 regulates reactive oxygen species levels and oxidative stress in radial glia progenitors during cortical development in mouse. Overall design: Ventricular tissue were isolated from three pairs of wild-type (WT) and Prdm16 cKO (conditional knockout) mutant E11.5, E13.5, E15.5, and E17.5 embryos. RNA was isolated using Qiagen RNeasy Micro Kit (QIAGEN). After RiboGreen quantification and quality control by Agilent BioAnalyzer, 125-150ng of total RNA underwent polyA selection and TruSeq library preparation according to instructions provided by Illumina (TruSeq Stranded mRNA LT Kit, catalog # RS-122-2102). Samples were barcoded and ran on the HiSeq 4000 platform, using the HiSeq 3000/4000 SBS Kit (Illumina).
