Lineage-specific methylation and transcriptomic patterns were identified in 6 primary immune cells from 24 healthy donors. We also look into the association of B cell-specific methylated sites with EBF1 ChIP-Seq binding patterns Overall design: Human blood was fractionated into 11 subsets using a combination of magnetic beads and flow cytometry, and DNA isolated from 1 – 2 million cells. A total of 300 ng genomic DNA was treated with sodium bisulfite using Zymo EZ-96 DNA Methylation Kit per the manufacturer's protocol, and each sample then hybridized to Illumina Infinium Human MethylationEPIC BeadChip arrays followed by processing and quantitation of methylation ( β ranging from 0-1) for each probe. Data from the Illumina Infinium MethylationEPIC array was analyzed in R using minfi package. noob-BMIQ normalization of the probes was performed after removing low quality probes (detection p values cutoff 0.01).
