Analysis of microbial diversity (protists) by eDNA amplicon sequencing in surface sediments of New Caledonian coastal ecosystems in relation to fluvial inputs influenced by catchment geology and mining activity. Surface (0-3cm) sediment samples were collected in march-april 2022. The V4-18S (TAReuk454FWD1/TAReukREV3 primers, Stoeck et al., Mol. Ecol., 2010) targeted protist communities. DNA extracts were amplified by PCR triplicate. Pooled samples were sent to the GeT-Biopuces platform (INSA, Toulouse, France) for Illumina MiSeq sequencing with a standard kit V3 (2x250 bp).
