MITF-dependent transcription has been implicated in processes of DNA replication and damage response (reviewed in Hou and Pavan, 2008; Cheli et al, 2010; Goding and Arnheiter, 2019). siRNA-depletion of MITF causes mitotic defects and the resulting accumulation of phosphorylated H2AX (gamma-H2AX) and TP53BP1 (also known as 53BP1) at sites of DNA damage (Giuliano et al, 2010; Strub et al, 2011). ChIP analysis in melanoma cells identifies a number of MITF target genes involved in the process of DNA replication and repair, including BRCA1, RAD53, RAD531L3, TERT and LIG1 (Strub et al, 2011). MITF binds to E-box elements in the promoter of the DNA ligase gene LIG1 to upregulate its expression. Consistent with this, the LIG gene shows enhanced H3K4 methylation and occupancy by RNA polymerase II (Strub et al, 2011).
