NFAT5 induction by the tumor microenvironment enforces CD8 T cell exhaustion [overexpression]

To decipher the molecular mechanism of NFAT5 or NFAT1 CA-RIT overexpressing CD8 T cells, we performed RNA sequencing on sorted P1A CD8 TILs seven days after CD8 T cell transfer. 24 hours activated P1A CD8 T cells were transduced with either empty-eGFP, NFAT5 or NFAT1 CA-RIT retroviral vectors and transferred 24h later into P511 mastocytoma bearing Rag1-/-B10D2 mice. Seven days post transfer CD8 TILs were sorted from the tumors and processed for RNAseq analysis.
