Bulk RNA sequencing of CD44Neg, CD44LoKitNeg, CD44LoKitPos and CD44High populations from mouse Aorta-Gonad-Mesonephros (AGM) region

The aim of the experiment was to compare newly defined CD44Neg, CD44LoKitNeg, CD44LoKitPos and CD44High populations from mouse Aorta-Gonad-Mesonephros (AGM) region Overall design: AGM from E9.5, E10 and E11 C57Bl/6 mouse embryos were isolated and single cell suspensions were generated. Cells were stained with the anti-VE-Cadherin, anti-CD44 and anti-Kit antibodies and 25 Vecadpos_CD44neg cells (n=10), 25 Vecadpos_CD44LoKitNeg cells (n=6), 25 Vecadpos_CD44LoKitPos (n=3) and 25 Vecadpos_CD44High cells (n=3) were sorted by flow cytometry. The SMART-Seq 2 method was then used to produce cDNA from these 22 samples. Sequencing libraries were generated and were later sequenced together on the Illumina Next Seq 500 sequencer.
