A knock-in mouse model has been generated where the serine residue at position 839 in the catalytic domain of receptor guanylyl cyclase C (product of the Gucy2c gene) has been converted to an isoleucine residue. Organoids were prepared from the small intestine of wild-type C57BL6N Tac mice and the mutant S839I mouse by established procedures. RNA was prepared from organoids and subjected to RNAseq analysis. In some cases, organoids were treated with interferon gamma (10ng/ml), or uroguanylin (1 micromolar) or both for 16h and then RNA prepared.d`
