Transcriptome gene expression analysis of root tip, leaf tip, shoot tip, root (without root tip) and leaf (without leaf tip) from Ceratopteris richardii sporophytes

To study genes specially expressed in root tip, leaf tip, shoot tip, root (without root tip) and leaf (without leaf tip) of Ceratopteris richardii, we carried out an RNA-seq to analyze gene expression levels from these five tissues. Overall design: Ceratopteris richardii (wild type RNWT1) spores were cultured on 1/2MS medium at 26 °C under a 16-h-light/8-h-dark photoperiod in a growth chamber. The root, leaf, root tip, leaf tip and shoot tip tissue were separated from 50 days sporophyte seedlings and frozen in liquid nitrogen for RNA extration. Library construction and deep sequencing were carried out using the Illumina HiSeq 3000 platform following the manufacturer's instructions by Genergy Biotechnology (Shanghai, China).
