This study uses RNA-sequencing of mouse melanoma derived tumor cells and microenvironmental EVs. B16F1 and B16F10 tumor cells were implanted subcutaneous in Black 6 mice. Formed tumors were digested enzymatically into a single cell suspension and cells and EVs (16.5k and 100k) isolated using differential ultracentrifugation. Total RNA was isolated using Trizol according to the manufacturers protocol and libraries prepared using Truseq RNA stranded ribo-zero library prep followed by sequencing on the Illumina NextSeq500 1x75bp High Output (300M) platform.
