The scale of introducing structural variants in vitro has been limiting for systematic studies of their effects. Novel CRISPR/Cas based technologies can perturb genomes with high efficiency and methods like CRISPR/Cas9, base editing and prime editing are part of a growing gene editing toolbox. The precision and efficacy of prime editing make it the current method of choice for systematic variant effect studies. Prime editing-based deletion approaches enable the systematic evaluation of structural variants in vitro and can help illuminate the role of non-coding DNA.
