Staphylococcus aureus strain Tx0117 is an MSSA with a type A beta-lactase that exhibits a high inoculum effect with cefazolin. After curing strain TX0117 of Bla using a combination of novobiocin and heat to create Tx0117, we found TX0117c to demonstrate an increased resistance to antimicrobial peptides. To further understand whether this phenotypic change was a direct result of beta-lactamase inactivation or whether the collateral mutations were selected by curing process, here we sequence and assemble the genome for the TX0117 and identify mutations between TX0117 and TX0117c. In addition to the curing mutation in BlaZ, the cured strain also showed mutations in five other genes including a truncation of relA, the GTP pyrophosphokinase involved in the stress response, and a substitution in mnA_1, which functions in the teichoic acid pathway.
