To identify transcriptomic signature of human airway epithelium as it undergoes full differentiation into mucociliated epithelial cells under air-liquid interface. We used microarrays to detail the global gene expression pattern from day 0 through day 28 following air-liquid interface in human airway epithelial cells and identified distinct clusters of gene expression during this process. In a time-course study, human small airway epithelial cells (hSAEpCs) were cultured in vitro and at successive points (days 0, 2, 4, 6, 8, 10, 14, 21 and 18 [labeled D0, D2, D4, and so forth] following air-liquid interface [ALI]) were lysed in situ, their RNA were extracted and hybridized on Affymetrix microarrays. Our objective here was to identify molecular signatures, as gene expression states for individual genes, that correlate with stage of hSAEpCs differentiation at various days post-ALI.
